چکیده

L( + )-Arginine hydrochloride was obtained from the Sigma Chemical Co., St Louis, Mo., U.S.A. The Arlco jackbean meal used in the preparation of urease was supplied by Hopkin and Williams Ltd., Chadwell Heath, Essex. The 'manganese-maleate solution' used in the preparation of arginase was made by dissolving 8-9 g. of MnSO4,4H20 and 1-8 g. of disodium maleate in glassdistilled water and making the volume up to 1 1. Buffers. 0-1 M-Glycine buffer was prepared by dissolving 7-5 g. of glycine in 200 ml. of water, adjusting the pH of the solution to 9 0 with 0-1N-NaOH and making the volume up to 11. with glass-distilled water. 3M-Acetate buffer, pH 4-6, was prepared by dissolving 408 g. of hydrated sodium acetate in 500 ml. of water, adding 170 ml. of acetic acid and making the volume up to 1 1. with glass-distilled water.

An Improved Method of Crystallization of Beef Liver Catalase

' .On the way of purification of beef liver arginase, Kitagawa and author') obtained a crystalline protein and ascertained it to be identical with the crystalline catalase which had been first isolated by Sumner and Dounce2) in 1937. In order to isolate the crystalline catalase, Sumner employed fractional precipitation by dioxane. An attempt to substitute acetone for dioxane was unsuccessful in...

Expression of human liver arginase in Escherichia coli. Purification and properties of the product.

Arginase is an enzyme that catalyses the hydrolysis of arginine to urea and ornithine. It is abundantly present in the liver of ureotelic animals (i.e. those whose excretion is characterized by the excretion of uric acid as the chief end-product of nitrogen metabolism), but its purification has hitherto not been simple, and the yield not high. Starting with a partially truncated cDNA for human ...

Molecular Characteristics of Chicken Liver Arginase By ENRICO GRAZI and ERMES MAGRI

A purification procedure for the preparation of chicken liver arginase in a homogeneous form is presented. The enzyme hydrolyses both arginine and argininic acid. Kinetic analysis reveals that the enzyme binds arginine when the amino group is protonated or unprotonated; however, the unprotonated form seems to be hydrolysed more rapidly. The enzyme exchanges with the medium approx. 1.6Mn2+ ions ...

Britain Purification and Properties of Arginase of Rat Kidney By GEORGE A . KAYSEN and HAROLD

L-Arginase from rat kidney was partially purified and some properties were compared with those of L-arginase of rat liver. The kidney enzyme was firmly bound to the mitochondrial fraction and after solubilization required arginine or an unknown factor in tissue extracts for stabilization after dialysis. The two enzymes differed also in stability with respect to acetone treatment, heating or fre...

The purification and properties of ox liver short-chain acyl-CoA dehydrogenase.

The FAD-containing short-chain acyl-CoA dehydrogenase was purified from ox liver mitochondria by using (NH4)2SO4 fractionation, DEAE-Sephadex A-50 and chromatofocusing on PBE 94 resin. The enzyme is a tetramer, with a native Mr of approx. 162 000 and a subunit Mr of 41 000. Short-chain acyl-CoA dehydrogenases are usually isolated in a green form. The chromatofocusing step in the purification pr...
